Associate Scientist- T Cell Engineering

SUMMARY:

An exciting opportunity for an immunologist is available in our ex vivo group; where we leverage top-notch CRISPR and other gene editing technologies to fully unleash the power of cell-based therapy for Autoimmune and Immuno-Oncology indications. The ideal candidate is versed in T cell immunology with a strong interest in T cell engineering and knowledge of T cell biology and related assays.
RESPONSIBILITIES:
- Responsible for functions related to the genome editing of T cells for cell therapies using CRISPR/Cas9.
- Conceptualize, design, execute, and troubleshoot experiments to knock out and insert genes within human T cells.
- Independently analyze results and optimize processes to drive towards higher editing efficiency and improved phenotype and viability.
- Perform in-vitro functional assays (cytotoxicity and cytokine release) to evaluate anti-tumor potency of engineered T cells.
- Collaborate with others across a matrixed organization to improve understanding of the biology of CRISPR/engineering of T cells.
- Execute scientific experiments, produce data, validate and optimize protocols for CRISPR/Cas9 genome editing.
- Act as liaison with the multiple teams in the ex-vivo emerging platform, ensuring alignment and support of T cell editing protocols for multiple programs.
- Analyze biological data and synthesize conclusions to help identify future directions of exploration.
- Maintain timely documentation of electronic lab notebooks and communicate /present data internally.
REQUIRED EXPERIENCE/SKILLS:
- Experience using CRISPR/Cas9 to edit genomes of primary human or mouse T cells
- Experience with multi-color flow cytometry of T cells with up to 6 markers, including intracellular staining methods
- Experience with in-vitro T cell assays including cytokine quantification (ELISA or MSD), and cell based cytotoxicity assays with tumor cell lines.
- Knowledge of cell therapy landscape and experience with CAR or TCR-T cell therapies is a plus
- Knowledge of the basic principles of cell biology and cell culture, including: maintenance of primary cells in tissue culture using sterile technique; passaging of cell lines; cell counting and replating; troubleshooting of cell culture issues; assessment of cell viability
- Diverse molecular biology techniques including DNA and RNA quantification through quantitative real-time PCR and single molecule digital droplet PCR
QUALIFICATIONS:
- Requires a Master’s degree along with at least two years of laboratory experience in industry.
- Alternatively candidates with a Bachelor’s degree in Biological Sciences, Bioengineering, Medical Biotechnology, or a directly related field plus at least four years of laboratory experience in industry will be considered.
